Title | "Novel Particle with Special Morphology: Design and Application in Vaccine Delivery" <Abstract> We have proposed a strategy to use a particle as a chassis, and then assemble antigen and other components on/in it to form a synthetic biomimetic vaccine. Particle can mimic the size and properties of pathogen. In this talk, we designed the particles with interesting morphology as the chassis to prepare synthetic vaccines. 1) We used nanoparticles instead of emulsifier to stabilize squalene oil to obtain Pickering emulsion, and then assembled antigen on the surface to form biomimetic vaccine. We found that Pickering emulsion chassis can mimic rough surface, deformability (pliability) and antigen mobility of pathogen. As a result, the vaccines showed excellent preventive effect and therapeutic effect. 2) We proposed to use a self-healing hollow particle (porous particle) as a chassis for inhaled vaccine. The large pores allowed easy encapsulation of nano-vaccine. And the micro-sized particle allowed efficient deposition of vaccine in lung. This micro-nano structured vaccine provided excellent systemic and mucosal immune response. |
