Title "Clickable Molecular Toolbox for Locoregional Fluorescent Imaging & ENCTACs Therapeutic Manipulation"

<Abstract>
Biological specificity arises from the precise and selective regulation of gene expression and protein interactions in response to intrinsic developmental programs and extrinsic signals. Comprehensive profiling of genetic expression and relevant protein interactions, accurate deciphering of the essential regulations involved, and importantly, real-time monitoring of their molecular biofunctions thereof demands unique and robust strategies with high sensitivity and fidelity that enable specific reporting of these biological events under complex and dynamic living environments.

Over the past decade, by integrating with chemical biology and molecular engineering strategies, our group has been actively involved in development of "smart' and unique molecular probes including near-infrared (NIR) dyes and lanthanide based fluorescent imaging contrast agents for real-time visualizing genetic expression, protein functions and cellular activities in various disease models. We have successfully established several sustainable imaging protocols that can not only realize high throughput (HTP) and flexibly screening of therapeutic candidates without the need of extensive chemical synthesis and effectively validating epigenetic oncogene deactivation, but can also precisely unravel the intrinsic therapeutic or resistant mechanisms in tumor and bacterial pathogens. Moreover, upon the biolabeling through unique enzyme responsive biorthogonal click conjugation, we have also developed a first-in-class ENCTACs (enzyme-derived clicking PROTACs) therapeutic strategy that can specifically respond to disease enzyme stimulation and selectively degrade unwanted proteins to re-modulate disease settings in vitro and in vivo. In this seminar, we will introduce all these latest discoveries. Our molecular imaging studies, fortified by unique chemical biology design and tissue penetrable molecular probes, provide invaluable insight into the realm of precision medicine and next-generation therapeutic development in the future.

Biography
Dr. Bengang XING received his Ph.D. degree from Nanjing University, China in Dec. 2000, after that he did his postdoctoral studies in HKUST, UCLA, and Stanford University. In early 2006, he moved to Singapore and started his independent career as Assistant Professor at NTU where he was promoted to tenured Associate Professor in 2011 and Full Professor in 2019. During 2011 and 2018, Professor Xing held concurrent appointments as Deputy Head in CBC and Assistant Chair in SPMS for academic education. Since 2022, Prof. Xing has been installed as a Full Professor in the School of Chemistry, Chemical Engineering and Biotechnology (CCEB) at NTU with his appointment as Assistant Chair for faculty development.

He has published over 150 peer-reviewed articles in highly regarded international journals and book chapters with a number of publications as highly cited articles in web of science, Thompson Reuters. He is also holding 20 international patents with some granted and licensed. By right, Dr. Xing has received numerous mainstream recognitions such as the Fellow of the Royal Society of Chemistry (FRSC) (2018), Minjiang Chair Professorship (2017), Young Innovator Award in Nano-Biotechnology (2017), top 2% global scientists in Chemistry, and Chairship of IUPAC/13th International Symposium on Bioorganic Chemistry (ISBOC13, 2023). He was also the guest editor for Advanced Drug Delivery Review (ADDR) in 2022. He is also currently serving as the Advisory Board Member of J. Control. Release, Bioconjugate Chemistry (ACS), Nano-theranostics, J. of Nanobiotechnology as well as Executive Associate Editor of Analytical Sciences etc.

Currently, his research interest is at the interface of chemical biology, molecular probe & imaging, bio-labeling as well as molecular therapeutics and nano-theranostics development., mainly focusing on the development of “smart” small molecules, peptides, and nanoprobes for biological process monitoring and diagnosis.
