Title | "Delineating the roles of lipid structures through metabolic manipulation of cells" <Abstract> Cellular membranes contain hundreds, if not thousands, of distinct lipid species. However, the current understanding about the relationship between lipid structures and functions remains severely limited. To overcome this, we are engineering cultured cells to manipulate their metabolism and alter their lipid composition. We use a highly efficient CRISPR-Cas9 approach to delete lipid metabolic genes in a multiplex manner, and combine it with lipidomics analyses to generate cells with altered lipid structures. With this, we found that the presence of very long-chain fatty acids in sphingolipids is critical to maintain overall membrane lipid composition of cells. In addition, we discovered the mechanism how cells accumulate polyunsaturated fatty acids at the inner leaflet of the plasma membrane, and found that this regulation is critical for ferroptotic cell death. |
