Title | "Two chemical stories, contributions in antiviral and bioluminescence research" <Abstract> This conference will describe the results of two research projects in which organic chemistry was combined with biology and biochemistry. - Alkoxypyrazole chemistry, from new chemical entities to original antivirals This project started with some chemistry of alkoxypyrazoles providing original accesses to new chemical entities. Then, following a screening of the resulting library, two compounds, modestly effective against the in cellulo replication of the RNA virus measles, were identified. The ensuing structure-activity relationship study led to a class of very efficient inhibitors of RNA virus replication in vivo as well as the identification of its target: the host cell dihydroorotic acid dehydrogenase (DHODH). - A light from the abyss, original contribution to luciferin chemistry and bioluminescence This project started with a required preparation of a large batch of furimazine, one of the best substrate of the luciferase NanoLuc/NanoKAZ which owes its origin to the luciferase of the bioluminescent shrimp Oplophorus Gracilirostris. Unexpectedly, our results soon led to a full-fledged study of the structure bioluminescence properties of this class of luciferin. We will describe these improvements as well as our chemical inputs in structural studies aiming at understanding how the catalyzed oxidation of this substrates, by NanoLuc as well Gaussia or Renilla luciferases, provides a photon. |
