Nelson (Adjunct Professor, Department of Entomology and Plant Pathology, Oklahoma State University , U.S.A.) ◆Title:"Virus-induced gene silencing: An in-depth evaluation of its power and limits for functional genomics studies” 〈Abstract〉 Virus-induced silencing (VIGS) is a powerful tool for gene function studies. For example, through VIGS the expression of an entire gene family or single genes within the gene family can be downregulated. The latter allows parsing of function among individual genes within a gene family for a particular activity. A multitude of other strengths associated with VIGS will also be presented and discussed. Although the benefits of the technology are extensive, its proper use is essential to a) avoid improper interpretation of results and b) achieve maximum silencing. For example, the improper construction of a host gene fragment for use in silencing can lead to unknowing silencing of related and even unrelated genes. The improper size and orientation of the host gene fragment in a particular VIGS vector can lead to suboptimal levels of target gene(s) silencing. The proper controls and host gene fragment design for VIGS studies will be presented here. Lastly, the worth of designing improved VIGS vectors for silencing will be discussed and an example of a recently improved VIGS vector will be presented. The power of this technology is truly extensive when properly implemented. |
